PT-141

Mechanism of Action & Benefits:

PT-141, also known as Bremelanotide, is a melanocortin-stimulating protein that has been found beneficial in the treatment of sexual dysfunctions such as erectile dysfunction in men and hypoactive sexual desire disorder in women.

Mechanism of Action

PT-141 works by binding to melanocortin receptors (MC3-R and MC4-R) in the hypothalamus in the brain. This activation of brain neurons has a direct erectogenic effect, resulting in on-demand erections. Unlike erectile dysfunction medications like Viagra and Cialis, which work through the vascular system to increase blood flow, PT-141 operates through the nervous system. As the receptors travel to the brain, PT-141 prompts them to send signals to the hypothalamus, increasing arousal, desire, and sexual satisfaction.

Benefits

The benefits of PT-141 are not limited to its erectogenic effects. It has been found to improve sexual desire in both men and women. It can also help improve sexual performance and elevate mood. The libido-enhancing effects of PT-141 typically last even longer than the direct erectogenic effects. Some sources also suggest that PT-141 may provide other benefits associated with regular exercise, such as reduced stress levels, and potential in reducing stress, regulating appetite and mood, and boosting the immune system.

Side Effects & Interactions:

The most commonly reported side effects include flushing, headaches, nausea, and irritation or redness at the injection site. Less commonly reported side effects include vomiting, blurred vision, dizziness, joint pain or swelling, runny or stuffy nose, darkening of the face or gums, and fatigue.

In some cases, PT-141 can cause an increase in blood pressure, which may lead to headaches, dizziness, or blurred vision. This effect usually subsides within 12 hours after the dose. However, if these symptoms persist for longer than 12 hours, it is recommended to seek medical attention.

PT-141 may also cause darkening of the skin on the face, gums, or breasts, especially in patients with darker skin and daily use. This skin change may be permanent, even after discontinuing the use of PT-141.

In terms of drug interactions, there is limited information available. One source suggests that the serum concentration of Naltrexone can be decreased when it is combined with Bremelanotide. However, it is important to consult with a healthcare provider for personalized advice on potential drug interactions.

Usage Guidelines:

Usage Guidelines for Subq Administration

  • Dosage: The recommended dose for adults is 1.75 milligrams (mg) injected under the skin at least 45 minutes before sexual activity. Increase/Decrease in .25mg increments for desired effect.

  • Frequency: Do not use more than one dose of PT-141 within 24 hours and limit the use to no more than 8 doses per month.

Usage Guidelines for Nasal Administration

  • Dosage: The dosing for PT-141 nasal spray is typically 1 to 2 sprays per nostril, administered at least one hour before the desired effect. Increase dose by 1 spray until reach desired effect.

  • Frequency: It is recommended not to exceed one dose of PT-141 in 24 hours and no more than eight doses per month.

Opinions on Nasal Administration

  • Effectiveness: Nasal administration of PT-141 has been shown to be effective in increasing sexual desire and improving sexual dysfunction in clinical trials.

  • Side Effects: Side effects from PT-141 were generally more severe in its subcutaneous injection form than its nasal spray form, but both had rates of adverse reactions.

  • Preference: Some users may prefer nasal spray due to the non-invasive nature of administration compared to injections.

  • Research: Intranasal PT-141 research was halted by the FDA due to concerns over increased blood pressure, and only subcutaneous delivery has been approved for use in women with hypoactive sexual desire disorder (HSDD).
